Tenis začala hrát ve třech letech pod vedením svého otce. Od roku 2005 je jejím trenérem Jaroslav Bulant. Jejím nejoblíbenějším povrchem je antuka.
Do světa profesionálního tenisu vstoupila již jako čtrnáctiletá v roce 2001 prohrou v 1. kole na turnaji okruhu ITF v Plzni. V roce 2004 vyhrála dva turnaje ITF v Bergamu a Bastadu. Průlom v její kariéře znamenal rok 2005, kdy získala p
rvní dva tituly na okruhu WTA v Estorilu a Forest Hills, postoupila do finále v Hertogenboschi, kde podlehla ve třech setech Kláře Koukalové, a sezónu zakončila na 50. místě žebříčku WTA.
V lednu 2006 dosáhla dosud nejvýznamnějšího úspěchu v kariéře, když zvítězila na turnaji v Gold Coast. Po semifinálové účasti na turnaji v Amelia Island postoupila na 26. místo žebříčku, sezónu zakončila na 41. místě.
2007 Na Australian Open prošla přes obhájkyni loňského triumfu a světovou dvojku Francouzku Mauresmovou do čtvrtfinále grandslamového turnaje. V ryze českém čtvrtfinále Australian Open podlehla Vaidišové. Senzační tažení Lucie Šafářové turnajem v Paříži skončilo těsně pod vrcholem. Česká tenistka vyhrála ve finálovém utkání s Petrovovou první set, ale víc už nedokázala. Předtím na turnaji Šafářová vyřadila tři hráčky z první světové desítky. Na French Open stejně jako na Australan Open porazila Mauresmovou. V osmifinále ale podlehla Rusce Chakvetadzeové. Na Wimbledonu si ve druhém kole obnovila zranění nohy, ale utkání proti Daniilidouvé vítězně dokončila. V dalším kole jí čekala favorizovaná Jankovičová. Ve Fed-cupu dokázala porazit slovenskou jedničku Hantuchovou a i díky tomuto vítězství se Češky dostaly do baráže o světovou skupinu. Tam narazily při neúčasti jedničky Vaidišové na Španělky. Šafářová se stala českou jedničkou, ale dokázala vyhrát jen jeden zápas a tak si musí Češky zase rok počkat.
Na Australian Open 2008 vypadla i kvůli zdravotním problémům hned v prvním kole, stejně jako v US Open a Wimbledonu. Jedině na French Open se dokázala dostat do druhého kola. Před US OPen vyhrála titul ve Forest Hill. Další rok se jí dařilo o něco lépe. Na AO se dostala do třetího kola, na FO vypadla ve druhém kole s V. Williamsovou. Na Wimbledonu a UO skončila již v prvním kole. V Quebecku prohrála až ve finále a další turnajový triumf nepřidala.
V roce 2010 na AO skončila v prvním kole na raketě V. Williams. Chuť si spravila na turnaji v Paříži, kde porazila například Peerovou, Schiavoneovou a Penneteovou. Ve finále pak podlehla Dementevové. Na FO se dostala do druhého kola a zbylé dva grand-slamové turnaje dohrála již v prvním kole.
Rok 2011 začala účastí ve čtvrtfinále v Brisbane. Na AO jí porazila ve třetím kole Zvonareva. Ve Fed Cupu proti Slovensku porazila lehce ve dvou setech Hantuchovou a pomohla týmu k postupu do semifinále. Na French Open vypadla ve druhém kole s Goregesovou. Na turnaji v Kodani si zahrála ve finále, ve kterém však podlehla světové jedničce Wozniacke. Na Wimbledonu v prvním kole přehrála krajanku Hradeckou, v tom druhém však podlehla Zakopalové. Na US Open vypadla ve třetím kole s Niculescovou.
Vrcholem sezony bylo bezesporu finále Fed Cupu. Lucka sice oba své zápasy prohrála, ale Petra Kvitová i deblistky Hradecká a Peschkehová nezaváhali a český tým se tak radoval s vítězství nad domácím výběrem Ruska.
